Effect of SUMO protease 1 on myeloma cell growth with lentivirus infection

  • 摘要: 目的 用含有SUMO蛋白酶1干涉载体的shRNA慢病毒感染骨髓瘤细胞系XG-7,探讨SUMO蛋白酶1对骨髓瘤细胞的影响作用,为临床治疗提供参考依据。方法 用含有SUMO蛋白酶1干涉载体的shRNA慢病毒和con慢病毒感染骨髓瘤细胞系XG-7,流式细胞仪和荧光显微镜观察骨髓瘤细胞系XG-7慢病毒感染效率,Q-PCR和western blot检测XG-7骨髓瘤细胞系感染shRNA和con病毒后SUMO蛋白酶1的敲除效率。结果con病毒和shRNA病毒感染XG-7骨髓瘤细胞系48 h后,病毒感染率均>98.00%;shRNA组SUMO蛋白酶1 RNA和蛋白表达量、骨髓瘤细胞系XG-7存活细胞比例均显著低于con组(P<0.05)。结论 SUMO蛋白酶1敲除能够抑制骨髓瘤细胞增殖,促进凋亡,影响骨髓瘤细胞的细胞周期和集落形成。

     

    Abstract: OBJECTIVE ShRNA lentivirus containing SUMO protease 1 interferometer vector was used to infect myeloma cell line XG-7, so as to explore the influence of SUMO protease 1 on myeloma cells, and provide references for clinical treatment. METHODS ShRNA lentivirus containing SUMO protease 1 interferometer vector and con lentivirus were used to infect myeloma cell line XG-7, flow cytometry and fluorescence microscopy were used to observe myeloma cell line XG-7 lentivirus infection efficiency, Q-PCR and Western blot were used to detect the knockout efficiency of SUMO protease 1 in myeloma cell line XG-7 infected with shRNA virus and con virus. RESULTS The infection rates of XG-7 myeloma cell lines at 48h after con virus and shRNA virus infection were above 98%. RNA and protein expression levels of SUMO protease 1 and the myeloma cell line XG-7 cell survival rate of shRNA group were less than those of con group (P<0.05). CONCLUSION Knockout of SUMO protease 1 can inhibit myeloma cell proliferation, promote apoptosis, and influence cell cycle and colony formation of myeloma cells.
