Abstract:
OBJECTIVE ShRNA lentivirus containing SUMO protease 1 interferometer vector was used to infect myeloma cell line XG-7, so as to explore the influence of SUMO protease 1 on myeloma cells, and provide references for clinical treatment.
METHODS ShRNA lentivirus containing SUMO protease 1 interferometer vector and con lentivirus were used to infect myeloma cell line XG-7, flow cytometry and fluorescence microscopy were used to observe myeloma cell line XG-7 lentivirus infection efficiency, Q-PCR and Western blot were used to detect the knockout efficiency of SUMO protease 1 in myeloma cell line XG-7 infected with shRNA virus and con virus.
RESULTS The infection rates of XG-7 myeloma cell lines at 48h after con virus and shRNA virus infection were above 98%. RNA and protein expression levels of SUMO protease 1 and the myeloma cell line XG-7 cell survival rate of shRNA group were less than those of con group (
P<0.05).
CONCLUSION Knockout of SUMO protease 1 can inhibit myeloma cell proliferation, promote apoptosis, and influence cell cycle and colony formation of myeloma cells.
