黄连素对铜绿假单胞菌生物被膜和藻酸盐合成相关基因的体外抑制作用

Inhibition effect of Berberine on biofilm formation and alginate bio-synthesis associated gene of Pseudomonas aeruginosa in vitro

  • 摘要: 目的 探讨黄连素对铜绿假单胞菌藻酸盐合成系统的调控作用。方法 结晶紫染色法和扫描电镜法分别观察不同浓度黄连素对铜绿假单胞菌生物被膜合成的干预作用;逆转录多聚酶链式反应(RT-PCR)技术检测高、中、低浓度黄连素对铜绿假单胞菌藻酸盐合成系统algR、algD、lasR和rhlR基因表达的影响。结果 与空白对照组比较,黄连素高、中和低浓度处理组,在24、72和168 h各时间段生物被膜厚度显著降低,差异有统计学意义(P<0.05);扫描电镜下,高、中、低浓度黄连素组对铜绿假单胞菌生物被膜的成熟抑制作用明显;与空白对照组比较,高、中和低浓度黄连素组algR、algD、lasR和rhlR基因表达量均显著减低,差异有统计学意义(P<0.05)。结论 黄连素对铜绿假单胞菌生物被膜和藻酸盐合成具有负调控作用,呈剂量依赖关系,可能与抑制algR、algD、lasR和rhlR等的基因表达有关。

     

    Abstract: OBJECTIVE To explore the regulatory effect of berberine on biofilm and alginate associated gene expression of Pseudomonas aeruginosa. METHODS Crystal violet staining and scanning electron microscopy were used to observe the regulatory effects of berberine on the biofilm formation of P aeruginosa at different concentrations. RT-PCR was applied to detect the effects of berberine on algR, algD, lasR and rhlR gene expression contributor to biofilm formation and aliginate bio-synthesis in P aeruginosa at high, medium and low concentrations. RESULTS Compared with the control group, the biofilm thickness in the high,medium and low concentration treated group were significantly thinner at 24 h, 72 h and 168 h(P<0.05). The biofilm maturity in the high, medium and low concentration treated groups were significantly inhibited under scanning electron microscopy. The expression of algR,algD,lasR and rhlR genes were significantly decreased at high, medium and low concentrations of berberine(P<0.05). CONCLUSION Berberine exerted negative regulatory effect on the biofilm formation and alginate biosy-nthesis of P aeruginosa in a dose-dependent manner, which may be related to the inhibition of algR, algD, lasR and rhlR expression in P aeruginosa.
