Abstract:
OBJECTIVE To explore the effect of Fuzheng Qingre Tonglin Decoction on the biofilm of multi-resistant
Escherichia coli.
METHODS The ability of biofilm formation in 18 clinical multi-drug resistant
E coli strains was determined by crystal violet staining, and strong biofilm-forming bacteria were selected for use. A self-made circular epidural catheter was inserted into the bladder of the rat through the urethra, and strong biofilm-forming bacteria were injected to establish a biofilm model of multi-drug resistant
E. coli urinary tract infection in rats. The rats were randomly divided into the model groups, levofloxacin group, Fuzheng Qingre Tonglin Decoction group, levofloxacin + Fuzheng Qingre Tonglin Decoction group and blank control group. On the 5 th day after the model was established, drugs were intragastric administrated for 14 days. The urine of the rats was collected for bacterial culture, and the bacterial morphology and biofilm structure on the surface of the foreign bodies in each group were observed by scanning electron microscopy. The amount of biofilm produced was quantitatively compared in all groups by the integration method.
RESULTS There were 13 strains of multi-drug resistant
E. coli producing membranes, accounting for 72.22%, which included 3 strains of strong biofilm-forming bacteria, 4 strains of medium biofilm-forming bacteria and 6 strains of weak biofilm-forming bacteria. The rate of urine positive bacterial culture in the blank control group were 0, and that in the model group, Fuzheng Qingre Tonglin Decoction group, levofloxacin group, levofloxacin + Fuzheng Qingre Tonglin Decoction group were 100.00%, 66.67%, 66.67% and 37.50%, respectively. Biofilm scores in the Fuzheng Qingre Tonglin Decoction group, levofloxacin + Fuzheng Qingre Tonglin Decoction group were significantly different from that in the model group(
P<0.05).
CONCLUSION This experiment showed that the proportion of clinically isolated multi-drug-resistant
E. coli producing biofilm was relatively high, and Fuzheng Qingre Tonglin Decoction had the effect of clearing bacterial biofilm. The combined administration of Fuzheng Qingre Tonglin Decoction with levofloxacin had a good ability to remove the biofilm of multi-drug resistant
E. coli, and can kill free biofilm bacteria.
