miR-27a及miR-206在EV71感染手足口病患儿中的表达及意义

Expression and significance of miR-27a and miR-206 in EV71 infected children with hand-foot-mouth disease

  • 摘要: 目的 研究微小核糖核酸27a(miR-27a)、微小核糖核酸206(miR-206)在肠道病毒71型(EV71)感染手足口病患儿中的表达及意义。方法 选取2019年11月-2020年11月在遵义医科大学第三附属医院(遵义市第一人民医院)就诊的EV71感染手足口病患儿66例,作为EV71感染组,选取同时期在医院健康体检无感染健康儿童30名为对照组,所有研究对象均采集静脉血检测血清miR-27a、miR-206表达水平,分析血清miR-27a、miR-206表达水平与EV71感染手足口病的关系。结果 EV71感染组miR-27a、miR-206表达均低于对照组,危重型患儿miR-27a、miR-206表达均低于重型、普通型患儿,重型患儿miR-27a、miR-206表达低于普通型患儿,差异有统计学意义(P<0.05)。Logistic回归结果显示,miR-27a、miR-206、白细胞(WBC)、C-反应蛋白(CRP)是EV71感染手足口病的危险因素(P<0.05)。受试者工作特征曲线(ROC)显示miR-27a、miR-206预测EV71所致手足口病曲线下面积(AUC)分别为0.703、0.768。结论 EV71感染手足口病患儿血清miR-27a、miR-206表达水平显著下降,低水平miR-27a、miR-206是EV71感染手足口病患儿发病的风险预测因子。

     

    Abstract: OBJECTIVE To study the expression and significance of microRNA 27 a(miR-27 a) and microRNA 206(miR-206) in children with hand foot mouth disease(HFMD) infected by enterovirus 71(EV71). METHODS Total of 66 cases of EV71 infected children with HFMD in our hospital from Nov 2019 to Nov 2020 were recruited and divided into the EV71 infection group; 30 uninfected healthy children in the same period were in the control group. Venous blood was collected from all children to detect the expression levels of serum mir-27 a and miR-206, and the relationship between the expression levels of serum mir-27 a and miR-206 and EV71 infected HFMD was analyzed. RESULTS The expression of miR-27 a and miR-206 in the EV71 infection group was significantly lower than that in the control group; the expression of miR-27 a and miR-206 in critically ill children was significantly lower than those in severe and common children; the expression of miR-27 a and miR-206 in severe children was significantly lower than that in common children(P<0.05). Multivariate Logistic regression analysis showed that low expression of serum miR-27 a and miR-206, WBC and CRP were risk factors for HFMD caused by EV71(P<0.05). The area under the curves(AUCs) of miR-27 a and miR-206 for the prediction of HFMD were 0.703 and 0.768, respectively. CONCLUSION The expression levels of miR-27 a and miR-206 in serum of children with HFMD infected by EV71 significantly decreased. Low levels of miR-27 a and miR-206 were risk factors for the prediction of EV71 infected children with HFMD.
