Abstract:
OBJECTIVE To explore the protective effect of total flavonoids from Quzhiqiao((Fructus Aurantii)) on lung injury of asthma mice infected with respiratory syncytial virus(RSV) through nuclear factor-κB(NF-κB) signaling pathway.
METHODS Totally 36 female BALB/c mice were randomly divided into the normal group, the model group, the positive group, the high-dose total flavonoids of Quzhiqiao group, the medium-dose total flavonoids of Quzhiqiao group and the low-dose total flavonoids of Quzhiqiao group, with 6 mice in each group. The asthma models were established by ovalbumin(VOA) and RSV solution. The reactivity of airway was observed, the expression levels of serum interleukin(IL)-4, IL-5 and IL-13 were detected with the use of enzyme-linked immunosorbent assay, the superoxide dismutase(SOD), malondialdehyde(MDA) and glutathione peroxidase(GSH-Px) in lung tissues were detected by biochemical method, Hematoxylin-eosin(HE) staining was performed for lung tissues, and the expression levels of NF-κB P65, nuclear factor inhibitor protein(IκB), IκB kinase(IκK) and caspase-3(Caspase-3) were detected by using western blot(WB).
RESULTS The levels of SOD and GSH-Px in lung tissues of the three total flavonoids of Quzhiqiao groups were significantly lower than those of the model group, the MDA level was reduced, significantly the expression levels of serum IL-4, IL-5 and IL-13 of the mice were reduced significantly(all
P<0.05).The expression levels of Caspase-3 and cytoplasmic IκBα, NF-κB P65 were significantly elevated in the three total flavonoids of Quzhiqiao groups and the positive groups(all
P<0.05).
CONCLUSION Total flavonoids from
Fructus aurantii has anti-inflammatory and anti-oxidative stress effect on the asthma mice with RSV infection, and the mechanisms may be related to the NF-κB signaling pathway.
