痰热清注射液对铜绿假单胞菌生物被膜形成的影响

Influence of Tanreqing injection on biofilm formation of Pseudomonas aeruginosa

  • 摘要: 目的 通过痰热清对铜绿假单胞菌生物被膜形成的影响的研究,为其引起的相关感染提供新的治疗途径。方法 采用2015年8-9月包头医学院第一附属医院细菌室提供的1株标准菌株ATCC27853,应用96孔培养板采用微量稀释法测定痰热清注射液对铜绿假单胞菌的最低抑菌浓度(MIC);MTT法测定痰热清注射液对铜绿假单胞菌生物被膜的最小抑膜浓度(SMIC);用扫描电镜观察药物对生物被膜的影响。结果 痰热清注射液对铜绿假单胞菌的MIC为100 μg/ml,随着痰热清注射液浓度的降低,其抗菌活性逐渐减小;扫描电镜下观察到50 μg/ml质量浓度的痰热清注射液对铜绿假单胞菌生物被膜的形成有显著影响。结论 痰热清注射液能有效抑制铜绿假单胞菌生物被膜的形成。

     

    Abstract: OBJECTIVE To study the influence of Tanreqing injection on biofilm formation of Pseudomonas aeruginosa so as to offer new treatment approaches for infections. METHODS From Aug 2015 to Sep 2015, the standard bacterial strain ATCC27853 provided by the bacteriology room of the First Affiliated Hospital of Baotou Medical College was adopted, and the minimum inhibitory concentration (MIC) of Tanreqing injection to the P.aeruginosa was determined by using 96 hole culture plate and micro dilution method, the SMIC of Tanreqing injection to the biofilm of P.aeruginosa was determined by means of MTT assay, and the influence of drugs on the biofilm was observed with the use of scanning electron microscope (SEM). RESULTS The MIC of the Tanreqing injection to the P.aeruginosa was 100 μg/ml, and the antibacterial activity was decreased with the reduction of concentration of the Tanreqing injection. The SEM observation revealed that the Tanreqing injection showed significant inhibitory effect on biofilm formation of the P.aeruginosa. CONCLUSION The Tanreqing injection can effectively inhibit the formation of biofilm of the P.aeruginosa.
