Association of CCND1 gene polymorphisms, Helicobacter pylori infection with susceptibility to gastric cancer

  • OBJECTIVE To observe the association of cyclin D1 (CCND1) gene polymorphism, Helicobacter pylori (Hp) infection with susceptibility to gastric cancer. METHODS Totally 82 gastric cancer patients and 70 chronic gastritis patients who were treated in the Seventh People's Hospital of Zhengzhou from Jan 2017 to Jan 2019 were respectively assigned as the gastric cancer group and the chronic gastritis group. The single nucleotide polymorphisms of CCND1 gene were detected by polymerase chain reaction-restriction fragment length polymorphism assay and direct sequencing for products. RESULTS The positive rate of Hp infection was 67.07% in the gastric cancer group, 60.00% in the chronic gastritis group, and there weas no significant difference. There were significant differences in the frequencies of AA, AG, GG genotypes and A, G alleles at rs9344 locus of CCND1 gene between the two groups (P<0.05). Logistic regression analysis showed that among the patients who were positive for Hp infection, there were significant differences in the frequencies of AA, AG, GG genotypes and A, G alleles at rs9344 locus of CCND1 gene between the two groups(P<0.05); among the patients who were negative for Hp infection, there were no significant differences in the frequencies of AA, AG, GG genotypes and A, G alleles at rs9344 locus of CCND1 gene between the two groups. Among the patients who were positive for Hp infection, there were significant differences in the frequencies of AA and AG plus GG genotypes at rs9344 locus of CCND1 gene between the two groups under recessive genetic model(P<0.05). There was significant difference in the 3-year survival rate between the positive Hp infection patients with AA genotype and the positive Hp infection patients with AA plus GG genotype(P<0.05).CONCLUSION The Hp infection and AA genotype at rs9344 locus of CCND1 gene are closely associated with the susceptibility to gastric cancer and prognosis. The positive Hp infection patients carrying with AA genotype have the increased susceptibility to gastric cancer, and the postoperative survival time is remarkably shortened.
