Association of IFNGR1 gene polymorphisms with prognosis of chronic gastritis adults complicated with Helicobacter pylori infection after eradication therapy

  • OBJECTIVE To explore the association of interferon-γ surface receptor complex (IFNGR1) gene polymorphisms with prognosis of chronic gastritis adults complicated with Helicobacter pylori (Hp) infection after eradication therapy. METHODS Totally 239 chronic gastritis adults complicated with Hp infection who were treated in the First Affiliated Hospital of Nanyang Medical College from Jan 2018 to Mar 2020 were recruited as the research subjects. All of the patients received the conventional quadruple Hp eradication program for treatment, with the course of treatment 10 days. The effect of the eradication therapy was evaluated after the treatment course for 4-6 weeks. The patients were divided into the eradication group with 136 cases and the no eradication group with 103 cases according to the effect of eradication. The clinical data were collected form the patients, the target fragments at rs1327475 locus of IFNGR1 gene were amplified by polymerase chain reaction, the serum specific Hp cytotoxin-associated gene A (CagA) antibody was detected by means of enzyme-linked immunosorbent assay, the virulence was determined by the critical value, and the reoccurrence of Hp gastritis was recorded. RESULTS There were significant differences in the overall distributions of genotypes and alleles at rs1327475 locus of IFNGR1 gene between the eradication group and the no eradication group(P<0.05); the frequencies of AA genotype and A allele of the no eradication group were higher than those of the eradication group(P<0.05). In the no eradication group, the reoccurrence rate of the patients carrying with IFNGR1 gen AA genotype was higher than that of the patients with carrying with GG/GA genotype(P<0.05). There were no significant differences in the Hp load and Hp virulence subtypes between the two groups. CONCLUSION The polymorphisms at rs1327475 locus of IFNGR1 gene remarkably affect the effect of eradication therapy among the population of chronic gastritis adults complicated with Hp infection in this area and have certain impact on the reoccurrence of disease.
