Expressions of JAK2/STAT3 signaling pathways in peripheral blood of children with rotavirus infectious enteritis and their clinical significance

Abstract
OBJECTIVE To explore the expressions of 1,25-Dihydroxyvitamin D3,(1,25-(OH)2 VitD3), CD11b, JANUS kinase 2 (JAK2)/signal transducer and activator of transcription 3 (STAT3) signaling pathways in peripheral blood of children with rotavirus infectious enteritis and analyze the clinical significance. METHODS A total of 127 children with rotavirus infectious enteritis who were treated in Jiangjin Maternal and Child Health Hospital from Jan 2020 to Jan 2021 were assigned as the case group, meanwhile, 132 healthy children who received physical examination were chosen as the control group. The levels of serum 1,25-(OH)2 VitD3. C-reactive proteome(CRP), procalcitonin (PCT), immunoglobulin(Ig)M, IgA, peripheral blood CD11b、JAK2 mRNA, STAT3 mRNA and CD4+/CD8+ were detected for the two groups of research subjects, and the correlation between the above indexes was observed. RESULTS The frequency of defecation, body temperature, peripheral blood CD11b, JAK2 mRNA, STAT3 mRNA, serum CRP and PCT were higher in the case group than in the control group (P<0.05); the serum 1,25-(OH)2 VitD3, IgM, IgA and peripheral blood CD4+/CD8+ were lower in the case group than in the control group(P<0.05). The children of the case group were grouped according to illness condition, the serum 1,25-(OH)2 VitD3 level of the severe group was lower than that of the moderate group and the mild group, and the serum 1,25-(OH)2 VitD3 level of the moderate group was lower than that of the mild group(P<0.05); the expression levels of peripheral blood CD11b, JAK2 mRNA and STAT3 mRNA of the severe group were higher than those of the moderate group and the mild group, and the expression level of above indexes of the moderate group were higher than those of the mild group(P<0.05). There was correlation between the above indexes(P<0.05). CONCLUSION The children with rotavirus infectious enteritis show low expression level of serum 1,25-(OH)2 VitD3, which declines with the aggravation of illness condition; the children show high expression levels of peripheral blood CD11b, JAK2 mRNA and STAT3 mRNA, which are elevated with the aggravation of illness condition. The four indexes may reflect the progression of inflammatory lesions and immune function.
